Study Purpose: This dataset examines the effect of chemogenetic activation of cholinergic and nitrergic myenteric neurons on motility of the colon.

Data Collection: Simultaneous tension recordings of the proximal (pC), middle (mC) and distal colon (dC) were collected.

Curator's Notes

Experimental Design: Transgenic mice expressing the chemogenetic proteins hM3Dq or hM4Di in cholinergic or nitrergic colonic myenteric neurons were used. The colon was dissected, rod placed within, three tension transducers glued to the proximal, middle, distal colon. Spontaneous contractions in all three channels were initially recorded. Then 10 micromolar Clozapine-N-Oxide (CNO) was added in order to activate the modified muscarinic receptors hM3Dq and hM4Di that were expressed in cholinergic and nitrergic neurons using ChAT-Cre and nNOS-CreERT2 drivers. When activated by CNO, these two modified receptors induce signaling through alpha-q or alpha-i pathways, leading to excitation or inhibition of neurons, respectively. These chemogenetic molecules are also referred to as designer receptors exclusively activated by designer drugs or DREAADs.

Subjects & Samples: This study used 24 mice from 6-18 weeks of age: 3 male and 3 female ChAT-Cre, conditional hM3Dq (for inducible Gaq signaling) mice or ChAT-Gaq mice 3 male and 3 female nNOS-CreERT2, conditional hM3Dq mice or nNOS-Gaq mice 3 male and 3 female ChAT-Cre, conditional hM4Di (for inducible Gai signaling) mice or ChAT-Gai mice 3 male and 3 female nNOS-CreERT2, conditional hM4Di mice or nNOS-Gai mice
